Analysis of Chloramphenicol in Shrimp Using Standard Addition Method Based on Diazotization
Penelitian ini bertujuan untuk mengetahui konsentrasi kloramfenikol pada udang menggunakan metode standar adisi berbasis reaksi diazotasi menggunakan serbuk Zn sebagai reduktor kloramfenikol, dilanjutkan dengan penggunaan N-(1-naftil) etilendiamin dihidroklorida sebagai zat pengkloping dan diukur pada panjang gelombang 565 nm. Berdasarkan pengujian, sampel udang ditemukan kandungan kloramfenikol sebesar 1964.91 mg/kg dan kandungan tersebut tidak memenuhi persyaratan yang ditetapkan European Commission yaitu sebesar 0.15 mg/kg. Nilai batas deteksi (LOD) sebesar 0.64 mg/mL dan nilai limit kuantisasi (LOQ) sebesar 0.64 mg/mL. Koefisien korelasi (R2) diperoleh sebesar 0.9991 pada rentang konsentrasi 0-50 ppm. Analisis hasil menunjukkan %recovery dalam analisis udang menggunakan metode ini sebesar 87.41-107.73% dengan rata-rata 109.38%.
This research aimed to determine the concentration in shrimp using the standard addition method based on diazotization reaztion using Zn powder as a reducing agent of chloramphenicol, followed by the use of N-(1-napthyl) ethylenediamine dihydrochloride as a coupling agent and measured at 565 nm. Based on the test, the shrimp sample was found to contain 1964.92 mg/kg of chloramphenicol and it exceeded the requirements set by the European Commission which was 0,15 mg/kg. The limit of detection (LOD) value is 0.19 mg/mL and the limit of quantitation (LOQ) value is 0.64 mg/mL. The correlation coefficient (R2) was 0.9991 for the concentration range of 0-50 ppm. The analysis of the results showed that the %recovery in shrimp analysis using this method was 97-41%-107.73% with an average of 109.38%.
