Umbi yakon (Smallanthus sonchifolius) mengandung senyawa aktif antara lain phenol, chlorogenic, ferulic, fruktooligosakarida, flavonoid, alkaloid, saponin, tanin, glikosida, dan steroid. Tujuan penelitian untuk mengetahui pengaruh variasi lama penyimpanan umbi yakon terhadap kadar senyawa flavonoid dan kemampuan toksisitas. Digunakan variasi lama penyimpanan 1, 6, dan 13 hari. Isolasi senyawa flavonoid melalui metode maserasi dengan pelarut etanol. Hasil isolasi senyawa flavonoid digunakan untuk uji kadar senyawa flavonoid dan kemampuan toksisitas. Hasil uji kadar senyawa flavonoid menggunakan spektrofotometri UV-Vis dengan metode kolorimetri, larutan standar kuersetin panjang gelombang 435 nm. Nilai kadar senyawa flavonoid pada lama penyimpanan 1,6 dan 13 hari secara berurutan sebesar 1,9617 ppm, 1,8917 ppm, dan 1,5179 ppm. Hasil uji kemampuan toksisitas menggunakan metode Brine Shrimp Lethlity Test (BSLT) dengan hewan coba larva udang Artemia salina Leach pada lama penyimpanan 1,6, dan 13 hari dengan nilai LC50 secara berurutan sebesar 76,968 µg/mL, 57,010 µg/mL, dan 53,188 µg/mL. Kesimpulan kadar senyawa flavonoid selama penyimpanan mengalami penurunan dan kemampuan toksisitas dari senyawa flavonoid memiliki sifat toksik.

Yakon tubers (Smallanthus sonchifolius) contain active compounds such as phenol, chlorogenic, ferulic, fructooligosakarida, flavonoids, alkaloids, saponins, tannins, glycosides, and steroids. The purpose of the study was to determine the effect of variations in storage of yakon tubers on the levels of flavonoids and toxicity. Variation storage time of 1, 6, and 13 days. Isolation of flavonoid compounds through maceration with ethanol solvents. The results of flavonoid compounds isolation were used to test the levels of flavonoids and toxicity. The test result of flavonoid compounds using UV-Vis spectrophotometry with colorimetric method showed the levels of flavonoid compounds at 1, 6, and 13 days of storage time respectively at 1.9617 ppm; 1,8917 ppm; and 1,5179 ppm. The toxicity ability test result using the Brine Shrimp Lethlity Test (BSLT) method with animals shrimp Artemia salina Leach larvae, at a storage time of 1, 6, and 13 days with sequential LC50 values of 76.968 µg/mL, 57.010 µg/ mL, and 53,188 µg/mL. The conclusion the levels of flavonoid coumponds during storage has decreased and the toxicity of flavonoids has toxic properties.
