Inhibitory Test of Papaya Leaf Extract (Carica papaya L.) Against the Growth Staphylococcus aureus Bacteria In Vitro
Penyakit mastitis yang disebabkan oleh bakteri Staphylococcus aureus banyak dijumpai pada hewan ternak. Daun pepaya memiliki kandungan senyawa aktif sebagai antibakteri yang banyak digunakan sebagai obat tradisional berbagai penyakit. Tujuan dari penelitian ini untuk mengetahui daya hambat konsentrasi ekstrak daun pepaya (Carica papaya L.) terhadap pertumbuhan bakteri S. aureus. Pembuatan ekstrak daun pepaya menggunakan metode maserasi dengan pelarut etanol 96%. Uji daya hambat menggunakan metode difusi sumuran. Konsentrasi ekstrak daun pepaya (C. papaya L.) yang digunakan yaitu konsentrasi 15%, 30%, 45%, dan 60%. Parameter penelitian ini yaitu diameter zona hambat pertumbuhan bakteri S. aureus. Data diameter zona hambat bakteri S. aureus dianalisis dengan menggunakan uji ANOVA. Hasil penelitian ini menunjukkan bahwa konsentrasi 30%, 45%, dan 60% ekstrak daun pepaya mampu menghambat pertumbuhan bakteri S. aureus dengan rata-rata diameter sebesar 7,667 mm, 11,333 mm, dan 16,333 mm, namun konsentrasi 15% belum mampu menghambat pertumbuhan bakteri S. aureus. Berdasarkan hasil penelitian menunjukkan bahwa ekstrak daun pepaya (C. papaya L.) bersifat antibakteri, sehingga perlu dilakukan penelitian lebih lanjut terkait pemanfaatan ekstrak daun pepaya (C. papaya L.) untuk pengobatan alternatif penyakit mastitis.
Mastitis caused by Staphylococcus aureus bacteria is often found in livestock. Papaya leaves contain active compounds as antibacterial which are widely used as traditional medicine for various diseases. The purpose of this study was to determine the inhibition of the concentration of papaya leaf extract (Carica papaya L.) on the growth of S. aureus bacteria. Making papaya leaf extract using maceration method with 96% ethanol solvent. Inhibition test using well diffusion method. The concentrations of papaya leaf extract (C. papaya L.) used were concentrations of 15%, 30%, 45%, and 60%. The parameter of this research is the diameter of the inhibition zone for the growth of S. aureus bacteria. The diameter of the inhibition zone of S. aureus bacteria was analyzed using ANOVA test. The results of this study showed that concentrations of 30%, 45%, and 60% of papaya leaf extract were able to inhibit the growth of S. aureus bacteria with an average diameter of 7,667 mm, 11.333 mm, and 16,333 mm, respectively, but a concentration of 15% had not been able to inhibit the growth of bacteria S. aureus. Based on the results of the study, it showed that papaya leaf extract (C. papaya L.) has antibacterial properties, so further research was needed regarding the use of papaya leaf extract (C. papaya L.) for alternative treatment of mastitis.
