Penelitian ini bertujuan untuk menganaisis pengaruh ekstrak etanol daun bandotan (Ageratum conyzoides L.) terhadap profil kadar gula puasa dan pulau Langerhans mencit induksi Alloxan monohydrate. Penelitian ini bersifat eksperimental dengan Rancangan Acak Lengkap (RAL). Hewan coba yang digunakan sebanyak 24 ekor mencit jantan galur BALB/c yang dibagi menjadi 6 kelompok, yaitu kontrol negatif (KN), kontrol positif (KP), dosis 28 mg/kg BB (DOS I), dosis 70 mg/kg BB (DOS II), dosis 112 mg/kg BB (DOS III), dan kelompok glibenclamide (GLB). Induksi Alloxan monohydrate dilakukan dengan injeksi intraperitoneal dan pemberian ekstrak etanol daun bandotan dilakukan secara per oral pada mencit diabetik. Kadar glukosa darah puasa diukur pada sebelum induksi aloksan (H-1), sesudah induksi aloksan (Hari ke-0), setelah perlakuan (Hari ke-7 dan Hari ke-14). Pengambilan pankreas dilakukan pada hari ke-14 dan pembuatan preparat dilakukan dengan pewarnaan HE. Hasil dianalisis menggunakan uji Kolmogorov-smirnov dan uji homogenitas. Kadar gula darah puasa dianalisis dengan uji Anova dan uji Duncan. Diameter pulau Langerhans dianalisis dengan uji Kruskall-walis dan uji Mann-Whitney. Hasil penelitian menunjukkan rata-rata selisih kadar glukosa darah pada KN, KP, DOS I, DOS II. DOS III, dan GLB berturut-turut sebesar 1,75; 18,25; -34,50; -55,00; -104,25; dan -24,75 mg/dL serta rearata diameter Pulau Langerhans kelompok KN, KP, DOS I, DOS II, DOS III, dan GLB berturut-turut sebesar 71,25; 19,87; 27,87; 46,50; 46,37; dan 25,87 µm. Berdasarkan data yang diperoleh dapat disimpulkan bahwa ekstrak etanol daun bandotan dalam berbagai dosis berpengaruh terhadap pulau Langerhans dan profil kadar gula darah puasa mencit induksi Alloxan monohydrate.

This research aimed to analyze the effect of ethanolic extract of bandotan leaves on fasting blood glucose level and Langerhans’s Islet profiles of Alloxan monohydrate induced mice. This was an experimental research with Completely Randomized Design (CRD). Animals used were 24 male mice of BALB/c strain were devided into 6 groups, namely KN (normal control), KP (positive control), DOS I (extract dose of 28 mg/kg BW), DOS II (extract dose of 70 mg/kg BW), DOS III (extract dose of 112 mg/kg BW), and GLB (glibenclamide treatment). Induction of Alloxan monohydrate was conducted by intraperitoneal injection and the bandotan leaf extract was administrated orally to diabetic mice. Fasting blood glucose level was measured before induction of Alloxan (H-1), after induction of Alloxan (Day-0), after extract treatments (Day-7 and Day-14). The pancreatic organ was taken on Day-14 and histophatological preparations was colored by HE staining. The results were analyzed using Kolmogorov-smirnov test and homogenety test. Blood glucose levels data were analyzed using Anova test and Duncan test. Langerhans’s islet diameter data was analyzed using Kruskall-walis test and Mann-Whitney test. The result showed changing of fasting blood glucose on KN, KP, DOS I, DOS II, DOS II, and GLB were 1,75; 18,25; -34,50; -55,00; -104,25; and -24,75 mg/dL respectively. The result of Langerhans’s Islet diameter on KN, KP, DOS I, DOS II, DOS II, and GLB were 71,25; 19,87; 27,87; 46,50; 46,37; dan 25,87 µm. Based on the data obtained it can be concluded that various doses of bandotan leaf extract influenced the profil of blood glucose levels of fasting mice and Langerhans’s Iset diameter on Alloxan monohydrate induced mice.
