Penyimpanan semen pada suhu rendah dapat menyebabkan penurunan kualitas spermatozoa salah satunya akibat ROS (Reactive Oxygen Species), maka dari itu dibutuhkan pengencer dengan antioksidan untuk melindungi spermatozoa. Antioksidan dapat diperoleh salah satunya dari Ekstrak Kulit Buah Alpukat (EKBA). Penelitian ini bertujuan untuk mengetahui pengaruh penambahan ekstrak kulit buah alpukat, serta memperoleh konsentrasi ekstrak kulit buah alpukat terbaik dalam mempertahankan kualitas spermatozoa sapi Simmental pada suhu 4-5°C. Desain penelitian yaitu Rancangan Acak Lengkap dengan 5 perlakuan pemberian konsentrasi EKBA 0%; 1%; 2%; 3%; dan 4% dengan pengulangan 4 kali. Pengamatan motilitas individu spermatozoa dilakukan di bawah mikroskop cahaya perbesaran 400x. Pengamatan viabilitas spermatozoa dengan metode pewarnaan eosin-negrosin dan diamati di bawah mikroskop perbesaran 400x. Pengamatan integritas membran sel spermatozoa menggunakan larutan Hypo-Osmotic Swelling Test (HOST) dan diamati di bawah mikroskop perbesaran 400x. Data dianalisis menggunakan uji Anova satu arah dan dilanjutkan uji Duncan. Hasil penelitian menunjukkan bahwa terdapat pengaruh signifikan penambahan EKBA terhadap kualitas spermatozoa sapi Simmental (P<0,05). Perlakuan EKBA 3% menunjukkan hasil motilitas 46,25 ± 1,85%, viabilitas 77,82% ± 0,90%, dan integritas membran sel spermatozoa 78,76 ± 1,30% yang optimal dibandingkan perlakuan lainnya. Berdasarkan hasil penelitian dapat disimpulkan bahwa penambahan EKBA 3% pada pengencer CEP memiliki nilai tertinggi dan konsentrasi paling optimal dalam mempertahankan kualitas spermatozoa dalam penyimpanan suhu 4-5°C selama 4 hari.

The storing semen at low temperatures could decrease in the quality of spermatozoa, one of which is due to ROS (Reactive Oxygen Species), therefore a diluent with antioxidants is needed to protect spermatozoa. Antioxidants can be obtained from natural materials, one of which is Avocado Fruit Peel Extract (EKBA). This study aims to determine the effect of adding avocado peel extract, as well as obtaining the best avocado peel extract concentration in maintaining the sperm quality of Simmental Bull at 4-5°C. The research design used was a completely randomized design with 5 treatments giving 0% EKBA concentration; 1%; 2%; 3%; and 4% with 4 repetitions. The Motility of individual spermatozoa observations was carried out with a 400x magnification under a light microscope. The Viability of spermatozoa was observed using eosin-negrosin staining method and observed with a 400x magnification under a microscope. The membrane integrity of the spermatozoa cell observation with Hypo-Osmotic Swelling Test (HOST) fluid and observed with a 400x magnification under a microscope. The data were analyzed used a one-way ANOVA test and continued with the Duncan's test. The result showed that the addition of EKBA significant affected the sperm quality of Simmental Bull (P<0.05). The treatment of 3% EKBA showed a motility result of 46.25 ± 1.85%, viability of 77.82% ± 0.90%, and membrane integrity of 78.76 ± 1.30% which was optimal compared to other treatments. Based on the results study, it can concluded that the addition of 3% EKBA in CEP diluent has the highest value and the most optimal concentration in maintaining the quality of spermatozoa in storage at 4-5°C for 4 day.
