Pengaruh Pemberian Ekstrak Holothuria leucospilota terhadap Aktivitas Fagositosis Sel Makrofag pada Mencit (Mus musculus) yang Diinduksi Alkohol

The Effect of Holothuria leucospilota Extract on Macrophage Cell Phagocytic Activity in Mice (Mus musculus) Induced by Alcohol

Ekstrak Holothuria leucospilota telah dilaporkan berpotensi dalam merangsang respons imun. Penelitian ini memiliki tujuan untuk mengetahui pengaruh pemberian ekstrak Holothuria leucospilota terhadap aktivitas fagositosis makrofag mencit yang diinduksi alkohol dan mengidentifikasi dosis ekstrak yang optimal sebagai imunomodulator. Mencit jantan dikelompokkan dalam 6 kelompok (4 mencit setiap kelompok): yang terdiri dari kontrol positif, kontrol negatif, serta kelompok perlakuan ekstrak Holothuria leucospilota dosis 0,61; 1,03; 1,45; dan 1,87 mg/20 g BB. Minuman beralkohol diberikan selama 8 hari dan pemberian ekstrak Holothuria leucospilota diberikan sebanyak 3 kali sehari selama 12 hari. Mencit dibedah dan diambil cairan intraperitoneal satu hari setelah perlakuan terakhir. Suspensi yeast digunakan sebagai bahan uji aktivitas fagositosis makrofag. Pembuatan preparat apusan menggunakan carbol fuchsin dan crystal violet. Data yang didapatkan dianalisis dengan Uji Parametrik ANOVA lalu dilanjutkan dengan Uji Tukey. Berdasarkan hasil analisis statistik diketahui ekstrak Holothuria leucospilota berpengaruh terhadap aktivitas fagositosis sel makrofag, yang ditunjukkan dengan menggunakan Uji Parametrik ANOVA dengan hasil p < 0,05. Simpulan dari penelitian ini ialah aktivitas fagositosis sel makrofag mengalami peningkatan sesuai dengan pemberian ekstrak Holothuria leucospilota. Dosis eksrak Holothuria leucospilota yang paling optimum dalam meningkatkan aktivitas fagositosis sel makrofag ialah dosis tertinggi, yaitu dosis 1,87 mg/20 gBB.

Holothuria leucospilota extract has been reported to have the potential to stimulate an immune response. This research aimed to identify the effect of Holothuria leucospilota extract on macrophage activity in mice (Mus musculus) induced by alcohol and identify the optimal dose of extract as immunomodulator. The male mice were divided into 6 groups (4 mice each group) positive control, negative control, and group treatments of Holothuria leucospilota extract with dose of  0,61; 1,03; 1,45; and 1,87 mg/20 g/BW. Alcohol drinks were given for 8 days and Holothuria leucospilota extract was given 3 times each day for 12 days. Mice were sacrificed and macrophage taken out of the intraperitoneal on one day after the last treatment. The phagocytic activity of macropaghe was tested by yeast suspension. The assembly of smeared prepared slide used carbol fuchsin and crystal violet dye. The data were analyzed using the ANOVA parametric test and continued with the Tukey Test. Based on the results of statistical analysis, was known that Holothuria leucospilota affected the phagocytic activity of macrophages, which indicated by the ANOVA parametric test with a result of p < 0,05. The conclusion of this research is phagocytic activity of macrophages raised according to the dose of Holothuria leucospilota extract. The optimal dose of Holothuria leucospilota  to increase the phagocytic activity of macrophages with a dose of 1,87 mg/20 g BW.
