Aktivitas Proteolitik pada Enzim Protease dari Bakteri Rhizosphere Tanaman Kedelai (Glycine max. L) di Trenggalek 

The Proteolytic Activity on Protease Enzymes from Soybean Plant Rhizosphere (Glycine max L.) in Trenggalek

Bakteri proteolitik merupakan bakteri yang mampu menghidrolisis protein menjadi asam amino sederhana. Tujuan dari penelitian ini adalah untuk menentukan aktivitas proteolitik pada enzim protease dari isolat bakteri yang diisolasi dan dikarakterisasi dari rhizosphere tanaman kedelai (Glycine max. L) di lahan pertanian Trenggalek, Jawa Timur. Penelitian ini dilakukan secara deskriptif kuantitatif. Bakteri yang diperoleh sebanyak tujuh isolat diuji aktivitas proteolitik dengan menggunakan media selektif Skim Milk Agar (SMA) ditandai dengan adanya zona bening (halo zone) yang terbentuk di sekitar koloni kemudian dikarakterisasi. Uji aktivitas enzim protease dilakukan pada tiga isolat yang membentuk zona bening terbesar dengan menginkubasi pada media selektif Skim Milk Broth (SMB) selama 72 jam dengan mengukur aktivitas enzim protease menggunakan spektrofotometer pada panjang gelombang 660 nm dari hasil inkubasi ke- 0, 4, 24, 48, 72 jam. Hasil Indeks proteolitik dengan zona bening terbesar diperoleh pada tiga isolat IRK1; IRK4 dan IRK7 secara berurutan 2,54, 1,77, dan 1,80. Sedangkan Aktivitas enzim protease pada isolat IRK1, IRK4, dan IRK7 berturut-turut diperoleh hasil eksponensial pada inkubasi 24 jam sebesar 5,97 U/mL, 5,43 U/mL, dan 4,56 U/mL. Sehingga dapat diketahui pada masa inkubasi 24 jam nutrisi yang tersedia memungkinkan sel untuk melakukan pembelahan dan jumlah sel meningkat secara cepat dalam memproduksi enzim protease.

    Proteolytic bacteria are bacteria that able to hydrolyze protein into simpler amino acids. This study aimed to determine the proteolytic activity of the protease enzymes from bacteria that isolated and characterized from the soybean (Glycine max. L) rhizosphere in Trenggalek farmland, East Java. This research was a descriptive quantitative research. The seven isolates bacteria which is obtained were tested for proteolytic activity using Skim Milk Agar (SMA) selective media, that characterized by the presence of a clear zone (halo zone) that formed around the colony. The protease enzyme activity test was carried out on three isolates that formed the largest clear zone by incubating in Skim Milk Broth (SMB) selective media for 72 hours by measuring the protease enzyme activity using a spectrophotometer at a wavelength of 660 nm from the results of incubation 0, 4, 24 , 48, 72 hours. The results of the proteolytic index with the largest clear zone were obtained in three isolate IRK1 ; IRK4 and IRK7 respectively are 2.54, 1.77, and 1.80. While the protease enzyme activity in IRK1, IRK4, and IRK7 isolates respectively obtained exponential results at 24-hour incubation are 5.97 U / mL, 5.43 U / mL, and 4.56 U / mL. So it can be seen that during the 24-hour incubation period the available nutrients allow cells to divide and the number of cells increases rapidly in producing the protease enzyme.
