Pengaruh Konsentrasi Hormon Paklobutrazol Terhadap Pertumbuhan Biji Sintetis Anggrek Tebu (Grammatophyllum speciosum) Secara In Vitro

Anggrek tebu (Grammatophyllum speciosum) dikenal sebagai anggrek spesies yang memiliki ciri khas perawakan dan perbungaan yang besar. Namun, keberadaannya di alam mengalami kepunahan akibat kegiatan eksploitasi, fragmentasi lahan, dan adanya kendala dalam hal perbanyakan, sehingga diperlukan teknologi biji sintetis melalui penambahan retardan paklobutrazol sebagai solusinya. Tujuan penelitian yaitu untuk mendeskripsikan pengaruh konsentrasi hormon paklobutrazol terhadap pertumbuhan biji sintetis anggrek tebu secara in vitro. Metode penelitian ini adalah eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL). Variabel manipulasi berupa konsentrasi paklobutrazol (0 mg/l, 1 mg/l, 3 mg/l, 5 mg/l, 7 mg/l, dan 9 mg/l), menggunakan 4 kali pengulangan (setiap unit terdiri dari 10 biji sintetis). Variabel responnya berupa pertumbuhan biji sintetis anggrek tebu meliputi kondisi fisik di setiap tahap, persentase daya tembus, daya hidup, kontaminasi dan browning, data dianalisis secara deskriptif kuantitatif.  Hasil dari penelitian menunjukkan adanya pengaruh pemberian paklobutrazol dengan berbagai konsentrasi terhadap pertumbuhan biji sintetis G. speciosum. Paklobutrazol 9 mg/l adalah konsentrasi optimal dalam menghambat pertumbuhan biji sintetis G. speciosum dengan tingkat daya hidup tertinggi dan daya tembus, kontaminasi serta browning terendah saat tahap regenerasi.

Sugarcane orchid (Grammatophyllum speciosum) is known as an orchid species with large stature and inflorescent characteristic. However, its existence in nature is experiencing extinction due to exploitation, land fragmentation, and problems in propagation, so synthetic seed technology is needed through the addition of paclobutrazol retardants as a solution. This research aimed to describe the effect of paclobutrazol hormone for in vitro growth synthetic seed of sugarcane orchid. The method of reserach was experimental by using Complete Randomized Design (CRD). The manipulation variable was paclobutrazol concentration (0 mg/l, 1 mg/l, 3 mg/l, 5 mg/l, 7 mg/l, and 9 mg/l), using 4 repetitions (10 synthetic seeds in each units). The response variable was sugarcane orchid synthetic seed growth like physical condition on every stage, percentage of emergence rate, survival rate, contamination, and browning, data were analyzed descriptively quantitatively. The research indicated that there was effect of paclobutrazol hormone concentration for in vitro growth synthetic seed of G. speciosum. Paclobutrazol 9 mg/l is the optimal concentration which was able to inhibit growth synthetic seed of G. speciosum with the highest survival rate and the lowest emergence rate, contamination, and browning.
