INFLUENCE OF ROSELLA FLOWER FILTRATE (Hibiscus sabdariffa) IN EGG YELLOW TRIS DILUENT ON SPERMATOZOA QUALITY OF ETAWA GOATS AT -196℃ STORAGE TEMPERATURE
Pelestarian genetik ternak dapat dilakukan dengan pemanfaatan spermatozoa unggul melalui semen beku. Penambahan antioksidan dalam media pengencernya perlu dilakukan untuk mempertahankan kualitasnya. Tujuan dari penelitian untuk mengetahui pengaruh penambahan filtrat kelopak rosella (FR) dengan konsentrasi optimal terhadap kualitas spermatozoa kambing PE. Jenis penelitian ekperimental yang menggunakan rancangan acak lengkap (RAL) terdapat 4 perlakuan (FR 0,0%, 1,5%, 3,0% dan 4,5%) dengan 5 ulangan. Parameter yang diamati meliputi motilitas, viabilitas dan integritas membran sel spermatozoa. Analisis data menggunakan uji Anava satu arah. Hasil signifikan dilanjutkan dengan uji Duncan. Hasil uji Anava menunjukkan penambahan filtrat kelopak berpengaruh signifikan terhadap kualitas spermatozoa kambing PE (P<0,05). Penambahan filtrat kelopak rosella 3% merupakan konsentrasi terbaik yang dapat mempertahankan kualitas spermatozoa kambing PE pada suhu penyimpanan -196℃ denganrata-rata nilai motilitas 40,10±4,93%, viabilitas 57,82±5,71%, dan integritas membran sel spermatozoa 51,78±5,15% dibandingkan dengan perlakuan lainya. Simpulan dari penelitian ini penambahan filtrat kelopak rosella 3% dapat mempertahankan kualitas spermatozoa kambing PE pada suhu penyimpanan -196℃, dapat diterapkan untuk inseminasi buatan.

Genetic preservation of livestock can be done by utilize superior spermatozoa through frozen semen. The addition of antioxidants in the diluent media is necessary to maintain its quality. The aims of the studied were to analyze effect of the addition of rosella petal filtrate (FR) with optimal concentration on the quality of PE goat spermatozoa. This type of experimental research used completely randomized design (CRD) there were 4 treatments (FR 0.0%, 1.5%, 3.0% and 4.5%) with 5 replicates. Parameters observed included motility, viability and cell membrane integrity of spermatozoa. Data analysis used one-way Anava test. Significant results were followed by Duncan's test. Anova test results showed the addition of petal filtrate had a significant effect on the quality of PE goat spermatozoa (P<0.05). The addition of 3% rosella petal filtrate is the best concentration that can maintain the quality of PE goat spermatozoa at a storage temperature of -196 ℃ with an average motility value 40.10±4.93%, viability 57.82±5.71%, and spermatozoa cell membrane integrity 51.78±5.15% compared to other treatments. The conclusion this studies that the addition of 3% rosella petal filtrate can maintain the quality of PE goat spermatozoa at a storage temperature of -196℃, can be applied for artificial insemination.
