Uji Antagonis Bacillus subtilis dan Bacillus megaterium terhadap Pertumbuhan Cercospora sp. yang Diisolasi dari Nepenthes sp.

Cercospora sp. merupakan jamur penyebab penyakit pada tanaman salah satunya pada Nepenthes. Pengendalian secara umum yang dilakukan untuk megurangi penyebaran penyakit ini dengan menggunakan fungisida sintetis. Bacillus subtilis dan Bacillus megaterium merupakan bakteri antagonis yang bisa dimanfaatkan sebagai agensia pengendali fitopatogen. Tujuan dalam penelitian ini yakni untuk menganalisis kemampuan antagonis Bacillus subtilis, B. megaterium dan kombinasinya dalam menghambat pertumbuhan jamur Cercospora sp. Penelitian dilakukan di Laboratorium Mikrobiologi FMIPA Unesa pada bulan Februari – April 2020. Jenis penelitian yang dilakukan merupakan eksperimental dengan 5 perlakuan meliputi kontrol negatif, B. subtilis, B. megaterium, kombinasi B. subtilis dan B. megaterium serta kontrol positif Mancozeb.Pengujian antagonis terhadap jamur Cercospora sp. dilakukan dengan metode sumuran. Analisis data hasil penghambatan pertumbuhan jamur dilakukan dengan  ANOVA satu arah lalu uji Duncan.  Jamur Cercospora sp. diisolasi dari tanaman Nepenthes sp. yang menunjukkan gejala bintik merah. Jamur tersebut memiliki ciri morfologi koloni berbentuk melingkar dan bergelombang, berwarna putih, miselia seperti kapas, serta memiliki hifa dan konidia bersekat. Pemberian perlakuan B. subtilis, B. megaterium dan kombinasi menunjukkan kemampuan dalam menghambat pertumbuhan Cercospora sp. dengan nilai persentase hambatan masing-masing sebesar 17.234 ± 8.085%, 17.714 ± 8.180%, dan 14.770 ± 11.245%. Bakteri B. subtilis dan B. megaterium mampu menghambat pertumbuhan jamur Cercospora sp. yang diisolasi dari Nepenthes sp..

Cercospora sp. is  a fungus that causes diseases in plants, one of which is Nepenthes. The common control to reduce the spread of this disease by using synthetic fungicides.Bacillus subtilis and Bacillus megateriumare antagonistic bacteria that can be used asbiological agents in diseases control in plants. This reasearch aims to analyze the antagonistic ability of B. subtilis, B. megaterium and their combination in inhibitingthe growth of the Cercospora sp. This research was conducted at the Mycrobiology Laboratory of FMIPA Unesa in February – April 2020. The type of research was an experimental with 5 treatments including negative control, B. subtilis, B. megaterium, combination and positive control Mancozeb. Antagonistic testingof Cercospora sp.was carried out using the well method. Data analysis of the growth inhibition was using one way ANOVA then continued using Duncan test. Cercospora sp. isolated Nepenthes sp. that showed symptoms of red spots. The fungus has the colony morphological characteristics of a circular and wavy, white color, mycelia like cotton, and has septate hyphae and conidia. B. subtilis, B. megaterium and combination treatments show ability to inhibit the growth of Cercospora sp. with the value of the percentage of obstacles respectively 17,234 ± 8.085%, 17.714 ± 8.180%, and 14.770 ± 11.245%.  B. subtilis, B. megaterium and combination treatments can inhibit the growth of the fungus Cercospora sp. which isolated from Nepenthes sp..
