Penelitian identifikasi profil miskonsepsi siswa pada konsep-konsep dalam materi fotosintesis dan respirasi tumbuhan menggunakan three-tier multiple choice diagnostic test telah dilakukan dengan tujuan untuk mendeskripsikan profil miskonsepsi siswa dan faktor-faktor penyebab terjadinya miskonsepsi tersebut. Penelitian ini merupakan penelitian deskripsi kuantitatif, yaitu mengembangkan instrument three-tier test yang divalidasi oleh ahli, mengujikan soal, wawancara, analisis hasil penelitian menggunakan tabel kategori dan kriteria tingkat miskonsepsi, serta penarikan kesimpulan. Sasaran penelitian ini adalah siswa MAN 1 Lamongan kelas XII MIPA 2, MIPA 4, dan MIPA 7 sebanyak 78 siswa, satu guru mata pelajaran biologi serta bahan ajar. Hasil penelitian menunjukkan profil miskonsepsi terdiri dari siswa mengalami miskonsepsi sebesar 55,21%, siswa tidak paham konsep sebesar 29,32%, dan siswa paham konsep sebesar 15,47%. Siswa yang mengalami miskonsepsi diperoleh kriteria miskonsepsi tinggi sebesar 28,21%, miskonsepsi sedang sebesar 66,67%, dan miskonsepsi rendah sebesar 5,13%. Faktor penyebab miskonsepsi terdiri dari siswa, buku teks, media pembelajaran, guru, cara mengajar, dan konteks.

The research of identification students misconception profile on photosynthesis and plant respiration by using three-tier multiple choice diagnostic test has actually been performed with the purpose of describing student’s misconception profile and its causes. This research was quantitative descriptive research, was using three-tier test validated by expert, test question for the student, interviews, analysis research results with table of three tier test criteria and category, and conclusions. The research subjects were 78 students of MAN 1 Lamongan grade XII Science 2, Science 4, and Science 7, biology teacher and the learning material. The results showed that students’ misconceptions profile consists 55,21% of student have misconception, the percentage of students who didn’t understand the concept was 29,32%, and there were 15,47% students who had understood the concept. The percentage of students who were categorized as having higher misconception were 28,21%, middle misconceptions were 66,67%, and lower misconceptions were 5,13%. Misconceptions are caused by the students, textbooks, learning media, teacher, learning method, and context.
